肾母细胞瘤中p53、Bcl-2基因调控和细胞凋亡研究 被引量:4

摘要 目的 分析肾母细胞瘤细胞凋亡和p5 3、Bcl-2蛋白表达的相关性及其与肾母细胞瘤组织学类型的关系。方法 对 3 0例肾母细胞瘤用DNA末端标记 (TUNEL)方法检测细胞凋亡 ,免疫组化LSAB法检测p5 3、Bcl-2蛋白的表达。结果  3 0例肾母细胞瘤凋亡指数 (AI)为 1‰~ 412‰ ,平均 99 7‰ ;p5 3、Bcl -2的表达指数PEI、BEI分别为 0‰~ 3 0 8‰、1‰~ 3 65‰ ,平均值分别为 81 8‰、90 2‰。细胞凋亡和p5 3与肾母细胞瘤组织学类型有显著相关性 ,细胞凋亡与p5 3呈负相关 ,Bcl -2与组织学类型、细胞凋亡均无关。结论 p5 3在调节肾母细胞瘤细胞凋亡中可能起重要作用 ,细胞凋亡和p5 3突变可能与肾母细胞瘤的细胞分化、预后有关 ,PEI。 Objective To analyse the correlativity between the apoptosis and p53,Bcl-2 expression and the relationship among apoptosis,p53,Bcl-2 expression and the histological type of nephroblastoma.To investigate the roles of p53,Bcl-2 genes in regulating function and apoptosis in the oncogenesis and progression of nephroblastoma.Methods Test apoptosis by using TUNEL and test p53,Bcl-2 expression by using immunohistochemistry(LSAB) in sections of 30 paraffin specimens of nephroblastoma.Result The apoptosis index(AI) of 30 specimens range from 1‰ to 412‰,p53 and Bcl-2 expression index(PEI、BEI)are 0~308‰ and 1‰~365‰ respectively.Statistics show that apoptosis and p53 expression have significant relation with histological type of nephroblastoma,and apoptosis has negative correlation with p53 expression,while Bcl-2 has no relationship with histological type,apoptsis,and p53 expression.Conclusions p53 expression may play a more important role than Bcl-2 expression in regulating apoptosis in nephroblastoma.Apoptosis and p53 gene mutation have relation with cell differentiation,and aggression of nephroblastoma.PEI and AI tests may provide experimental basis of studying the generation and judging prognosis of nephroblastoma.
