游离铁在脊髓组织匀浆脂质过氧化中的作用

摘要 目的 观测游离铁在脊髓组织匀浆脂质过氧化中的作用。方法 SD大鼠脊髓组织匀浆中分别加入Ringer’s液、枸橼酸亚铁溶液 (10 μmol)或去铁胺溶液 (6 μmol) ,在 37℃恒温水浴中孵育 2h ,测定加入不同浓度Fe2 + 及去铁胺对脊髓组织匀浆MDA的影响。结果 脊髓匀浆混合液孵育 2h后 ,随着Fe2 + 浓度的增加MDA不断增加 ;当Fe2 + 浓度为 2 0 μmol时MDA浓度最高 ,随后逐步降低 ;加入去铁胺组与相同Fe2 + 浓度不加去铁胺组比较可明显减少MDA的产生。结论 游离铁能显著增强脊髓匀浆的脂质过氧化 。 Objective To observe the effects of free iron on the changes of lipid peroxidation in spinal cord homogenates in rats.Methods The Ringer's solution,Ferrous citrate (10 μmol) and deferoxamine mesylate(DFO)(6 μmol) were added separately to spinal cord homogenates of SD rats The mixture were incubated at 37°C in a shaking waterbath for 2 hours,and were determined the level of MDA.Results The level of MDA increased following the increase of Ferrous citrate,and reached highest level when Ferrous citrate was 2.0 μmol,then progressively decreased.DFO can decreased the formation of MDA at the same Ferrous citrate Conclusion The free iron can augment the lipid peroxidation in spinal cord homogenates.
