大孔树脂吸附法富集地锦草总黄酮的工艺研究 被引量:24

摘要 目的:研究大孔树脂富集地锦草中有效成分地锦草总黄酮的工艺条件及参数.方法:以地锦草总黄酮含量为考察指标,采用正交试验设计,选用L9(34)正交试验表,以直接影响地锦草总黄酮收率的上柱量、吸附时间及洗脱液的浓度为试验因素,每个因素取三个水平,进行地锦草总黄酮大孔树脂吸附法富集的工艺研究.结果:10ml样品液(每1 ml 75%乙醇液含地锦草干浸膏0.5 g)上柱,静置吸附时间30min,用95%乙醇洗脱地锦草总黄酮为最佳工艺.结论:通过大孔树脂富集,在洗脱液干燥后的总固物中地锦草总黄酮含量大于16%,高于醇提干浸膏的含量(7.61%),且洗脱率大于93%.采用此法可较好地富集地锦草中的有效成分. Objective: To study the optimal technology parameters of the enrichment process for total flavonoids of Euphorbia himifusa with macroporous resin.Methods:The enrichment process was studied by orthogonal experiment design using orthogonal form L9(34) with the content of total flavonoids from Euphorbia himifusa as indexes, and three factors were chosen in this experiment, including the saturation of the extract sicca from Euphorbia himifusa, duration of absortion and the consistence of alcohol as eluent. Results: The results showed that the optimal enrichment process was that 10ml of the extract of Euphorbia himifusa (i. e. 1ml 75% alcohol contains 0. 5g the extract sicca from Herba Euphor-biae Humifusae) was absorbed 30min with a column of macroporous resin (r 15 mm x H160mm) and the total flavonoids was eluted from the macroporous resin with 95% alcohol. Conclusion:The elutive rate of total flavonoids was above 93% by means of the macroporous resin. So this process enriching the total flavonoids of Euphorbia himifusa is feasible.
