摘要
目的 用酶标示踪、非荧光双标术观察和研究心脏和内关穴区的神经投射的同源性 ,为阐明针刺内关治疗、改善心功的神经网络机理提供依据。材料和方法 :健康雄SD大鼠 7只 ,每只动物在心前壁注入 3 0 %HRP ,同时在内关穴区注入霍乱原 β亚单位 (CB) ,存活 72小时 ,灌注、固定 ,取材 ,冰冻切片。用四甲基联苯胺 钼酸胺 (TMB AHM)法显示心脏神经投射的HRP阳性细胞 ;再用制备并标定效价的CB抗体作免疫组化染色 ,以显示内关穴区神经投射的CB阳性细胞 ;光镜观察HRP阳性、CB阳性和CB ,HRP双标阳性细胞。结果 :发现在C5 C7脊神经节和迷走神经节内有CB和HRP均为阳性的双标阳性神经元出现。结论 :以上结果首次证实心脏和内关穴区的神经纤维有部分来自脊神经节和颈迷走神经节中的同一个神经元 ,为解释心肌缺血时左臂“牵涉痛”
Objective:To study the role of non flourescence double labeling technique in homologus relationship of the neuroprojection between heart and Neiguan point area.Methods:SD rats were injected with HRP in the heart wall and with CB in the Neiguan point in the same time respectively.TMB AHM method was used to show HRP labeled neurons projected from heart and immunohistochemicl method to show CB labeled neurons projected from Neiguan point area.All the labeled neurons were observed under microscope.Results:The results show that the double labeled neurons could be seen in C 5 C 7 spinal ganglions and inferior vagus ganglion.The brown granules and blue granules occurred in the plasm of the same neuron.Conclusion:It can be drawn that some nerve fibers in the heart and the Neiguan point area originate from a same neuron in the spinal ganglions or the inferior vagus ganglion,which can explain the 'involved pain' during heart attack,providing a morphologic foundation for acupuncturing Neiguan point to treat heart diseases.
