NPCLC中P_(16)和Rb基因产物的表达情况及与细胞增殖的关系 被引量:1

摘要 目的:探讨抑癌基因的P16、Rb基因产物在非小细胞肺癌化S-P法,检测68例NPCLC 患者的手术切除标本的 P16、Rb的表达情况,并进行PCNA检测,计算细胞增殖指数(PI, proliferation index)。结果:在68例NPCLC 患者标本中,P16、Rb的阳性率分别为64.7%、88.5%,二者在不同组织类型的NPCLC中的表达无显著性差异(P>0.05);二者的表达均随PI分级的升高有不同程度的下降。31例低度恶性NPCLC肿瘤中,Rb均为阳性表达(+、++),29例P16阳性或强阳性(+、++)表达;在37例高度恶性NPCLC中29例Rb阳性或强阳表达,其中P16、表达缺失22例(75.9%);而在8例Rb阴性的NPCLC中有6例(75%)为P16阳性或强阳性表达。结论:1、P16及Rb参与NPCLC的细胞增殖过程,并且P16蛋白的缺失可能是NPCLC由低度恶性向高度恶性过渡重要事件;2、P16与Rb在高恶性NPCLC肿瘤中呈负相关性表达,这种负相关在NPCLC中是一常见事,是NPCLC高度恶性的标志之一。 Objective: To study expression of product of genes p16 and Rb in non parvicellular lung carcinoma (NPCLC), and relation ship between genes P16 and Rb and cell proliferation. Methods: Using S-P immunohistochemical methods, the expressions of P16\ Rb and PCNA protein in 68 cases of NPCLC were studied ,and the proliferation index(PI) was calculated. Results: The positive rates of P16 and Rb were 64.7%and 88.5% in 68 cases of NPCLC, with no significant difference in different histological types of NPCLC (P>0.05). The P16 protein expression was different in different PI grading, with the P16 protein expression lower and the PI grading higher, and the Rb expression was lower with the rising PI grading. The Rb expression was all positive ,and P16 showed positive expression in 29 of 31 low magilant NPCLC. The positive expressions of Rb and P16 were showed in 29 and 15 cases respectively in 37 high magilant NPCLC; Six showed positive or strong positive or strong positive expression in 8 cases of Rb negative expression. Conclusion: 1.P16 and Rb take part in process of cell proliferation of NPCLC, and the loss of P16 protein is important from low to high magilant changes in NPCLC . 2. P16 and Rb are negatively correlative expression in high magilant NPCLC, and this correlation is common and may be one magilant sign in NPCLC.
