失血性休克及复苏后肠组织能量代谢变化与细胞增殖的研究 被引量:2

Study of energy metabolic changes and cellular proliferation in intestine after hemorrhagic shock and resuscitation

摘要 目的 探讨肠组织在失血性休克及复苏后的能量代谢变化与上皮细胞增殖的相关性。 方法 Wistar大鼠 4 8只 ,分为对照组、单纯休克组和复苏 0 .5 ,1,2 ,6h组 ,每组 8只 ,采用休克复苏模型 ;取各组空、回、结肠组织 ,用高效液相色谱法测定ATP、ADP和AMP。另取Wistar大鼠6 4只 ,分为对照组、单纯休克组和复苏 2 ,6 ,12 ,2 4 ,72h和 7d组 ,每组 8只 ,制作休克复苏模型 ,采用3 H -TdR掺入法测定各组肠上皮细胞增殖能力。 结果 失血性休克后肠组织ATP含量显著降低 ,为对照组的 2 6 .3%~ 39.7% ;复苏后ATP恢复较慢 ,复苏 6h后为对照组的 6 3.5 %~ 6 7.7%。失血性休克及复苏早期 (2h)肠上皮增殖明显受抑制 ,复苏 12h显著升高 ,72h增殖达高峰 ,复苏后7d基本恢复正常。 结论 休克及复苏后肠上皮细胞增殖与能量代谢恢复基本一致。 Objective To find the correlation between energy metabolic changes and cellular proliferation of the intestine after hemorrhagic shock and resuscitation. Methods Forty eight Wistar rats were divided into six groups: control group, shock group, resuscitation 0.5, 1, 2 and 6 hours groups (n = 8 in each group). The specimens of the jejunum, ileum and colon were made using a hemorrhagic shock and resuscitation model. The contents of ATP, ADP and AMP were measured by HPLC. Other sixty four Wister rats were divided into eight groups: control group, shock group and resuscitation 2 , 6 , 12 , 24 , 72 hours and 7 days groups (n = 8 in each group). The capability of proliferation of the intestinal epithelium was measured by using 3HTdR. Results The ATP content of the intestinal tissue was significantly decreased after hemorrhagic shock, being 26.3% 39.7% of the control. After resuscitation, it was increased slowly and was 63.5%~67.7% of the control 6 hours after resuscitation. The proliferation of the intestinal epithelium was significantly suppressed during hemorrhagic shock and at the early stage (2 hours) after resuscitation. However, it was remarkably increased at 12 hours, peaked at 72 hours after resuscitation and returned to normal at 7 days. Conclusions The intestinal epithelial proliferation is in accordance with the energy recovery after hemorrhagic shock and resuscitation.
