摘要
为探讨大黄泻下作用与肠道 5 -HT及其受体的关系 ,将成年SD大鼠 2 0只随机分为灌服大黄组 (A组10只 )和对照组 (B组 10只 )。采用免疫组化和医学图像分析系统相结合的方法 ,测定各组大鼠十二指肠 5 -HT细胞的数目、面积、平均灰度、细胞内 5 -HT含量及 5 -HT受体的含量。结果发现A组大鼠十二指肠 5 -HT细胞的数目 (10 8 80± 5 2 6 )、面积 (30 18± 2 18) μm2 、5 -HT含量 (0 340 9± 0 0 30 8)AU、5 -HT受体含量 (0 3188± 0 0 312 )AU均高于B组 (P <0 0 1) ,平均灰度 (12 3 38± 3 94 )较B组降低 (P <0 0 1)。结果表明大黄的泻下作用可能与肠道 5 -HT及其受体含量的增高有密切关系。
In order to study the relationship between drastic effect of rhubarb and 5-HT and its receptor in duodenum tissues.Twenty SD adult rats were divided into two groups at random.group A:gastrogavaged with rhubarb group(n=10);group B:control group(n=10).The immunohistochemistry and MIPS were used to measure 5-HT cell number,square,mean grey level and the content of 5-HT and it′s receptor in their positive cells in every rat′s duodenum.In group A,rat duodenum 5-HT cell number(108 80±5 26),square(30 18±2 18)μm 2,the content of 5-HT(0 3409±0 0308)AU,the content of 5-HT receptor(0 3188±0 0312)AU were all higher than those in group B(P<0 01),mean grey level(123 38±3 94)was lower than that in group B(P<0 01).Rhubarb′s drastic effect was highly associated with the increase of 5-HT and 5-HT receptor′s content in dcodenum.
