摘要 脯氨酸的积累与植物对干旱和盐胁迫的抗性有关。P5CS基因编码一个双功能酶 ,具有谷氨酸激酶和谷氨酸 -γ -半醛脱氨酶活性 ,催化脯氨酸合成过程中的前 2步反应。它是脯氨酸合成的限制因子。用生物素标记的荧光原位杂交技术 ,以蛾豆 (Vignaaconitifolia)中克隆到的P5CS基因作探针对水稻进行物理作图。该探针定位于水稻第 9染色体的短臂近端点处 ,信号点距着丝粒的相对距离为 ( 5 1 .79± 1 .2 4 ) %。 Proline (Pro) accumulation has been associated with tolerance to drought and salinity stresses in plants. Δ ' pyrroline 5 carboxylate synthetase ( P5CS ) gene encoded a bifunctional enzyme with both g glutamyl kinase and glutamic g semialdehyde dehydrogenase activities that catalyzes the first two steps in proline biosynthesis. It was rate limiting for Pro synthesis. Using a biotin labeled fluorescent in situ hybridization (FISH) technique, P5CS cDNA clone from mothbean ( Vigna aconitifolia ) as a probe was approximately located onto the long arm of rice chromosome 9. The percentage distance (from the hybridization sites to centromeres) was ( 51.79± 1.24)%. P5CS is widely present in monocot and dicot.
