摘要
目的 探讨大肠癌组织端粒酶各组分基因 (hTR、TP1、hTERT)表达及其与端粒酶活性的关系。方法 分别采用原位逆转录聚合酶链反应和端粒重复序列扩增法 ,检测 46例大肠癌及其相应正常组织中的端粒酶各组分基因表达及端粒酶活性。结果 hTR或TP1mRNA在大肠癌与其相应正常组织中表达比较 ,差异无显著性 (P >0 .0 5 ) ,而hTERTmRNA在大肠癌组织中表达显著高于其相应正常组织 (P <0 .0 5 )。大肠癌组织端粒酶活性亦显著高于其相应正常组织端粒酶活性 (P <0 .0 5 )。端粒酶活性与hTERTmRNA表达显著相关 (r =0 .848,P <0 .0 5 ) ,而与hTR或TP1mRNA表达无相关性 (P >0 .0 5 )。结论 hTERTmRNA在端粒酶活性调节中可能起关键作用 。
Objective To explore the expression of telomerase subunits and its relationship with telomerase activity in colorectal cancer.Methods The expression of telomerase subunits (hTR,TP1,hTERT) and telomerase activity were detected in colorectal cancerous tissues and normal tissues from 46 patients by using in situ reverse transcription polymerase chain reaction and telomeric repeat amplification protocol,respectively.Results There was no statistically significant difference between the expression of hTR or TP1 mRNA in colorectal cancerous tissues and their expression in normal tissues (P>0.05).Whereas the difference of hTERT mRNA between cancer and normal tissues was significant (P< 0.05). Frequency of telomerase activity expression in cancerous tissues was much higher than that in normal tissues (P<0.05).There was a significant correlation between the expression of hTERT mRNA and telomerase activity (r=0.848,P<0.05).No correlation was observed between telomerase activity and the expression of hTR or TP1 mRNA(P>0.05).Conclusion hTERT mRNA may play a critical role in regulation of telomerase activity.The detection of hTERT mRNA may be a useful marker for diagnosis of colorectal cancer.
