摘要
目的 探讨尼莫地平对过氧化氢 (H2 O2 )诱导人血管内皮细胞凋亡的影响。方法 在人脐静脉血管内皮细胞培养基中分别加入 H2 O2或 H2 O2 +尼莫地平 ,培养 2 4 h,用流式细胞仪 PI标记法、原位末端脱氧核苷酸转移酶 (Td T)标记法 (TUNEL)和 HE染色法对细胞凋亡进行鉴定和检测。结果 H2 O2 能明显地诱导培养的人血管内皮细胞发生凋亡 ,尼莫地平可显著降低 H2 O2 诱导的内皮细胞凋亡率 (P<0 .0 1 )。结论 尼莫地平对 H2 O2 诱导的血管内皮细胞凋亡具有保护作用。
Objectives To study the effect of Nimodiping on vascular endothelial cell apoptosis induced by hydrogen peroxide (H_2O_2).Methods Cultured vascular endothelial cells (VECs) were treated with H_2O_2 or H_2O_2 + Nimodiping for 24 hours. Flow cytometer, terminal deoxynucleotidyl transferase-mediated dUTP nick endlabeling (TUNEL), HE staining were employed to monitor cell apoptosis. Results H_2O_2 was able to induce apoptosis in human VECs. Nimodiping could depress human VECs apoptosis. Conclusions Nimopdiping could protect VECs apoptosis induced by H_2O_2.
