摘要
为了研究不同地区栽培赤芝及野生赤芝中灵芝酸类成分含量的差异 ,建立了赤芝中灵芝酸类活性成分含量的反相高效液相色谱 ( RP-HPLC)测定方法。该方法采用 YWG-C1 8色谱柱 ( 2 50 mm× 4 .6 mm ID,1 0μm) ,甲醇 -水 -乙酸 ( 49.5∶ 4 9.5∶ 1 )为流动相 ,流速 1 .0 m L/ min,柱温为室温 ,检测波长 2 50 nm。结果 ,灵芝酸 A( ) ,灵芝酸 C( ) ,灵芝酸 D( ) ,灵芝酸 E( ) 4种成分的线性范围分别为 0 .0 98~ 3.1 38、0 .0 91~2 .91 3、0 .0 89~ 2 .850、0 .0 89~ 2 .850μg,方法灵敏、准确 ,重复性好 ,回收率分别为 93.93%、96 .83%、93.0 1 %、90 .4 0 %。栽培赤芝和野生赤芝子实体中 4种灵芝酸成分的总含量分别为 0 .799~ 3.555、1 .6 1 9~2 .36 0 mg/ g。
To study the difference of ganoderic acid contents in Ganoderma lucidum of the culture and wilding from different sources,a method for determination of content of ganoderic acid in Ganoderima lucidum was established by reversed phase high performance liquid chromatography. The analytical column used was 250 mm×4 6 mm ID, packed with YWG C 18 (10 μm). The mobile phase was a mixture of MeOH H 2O HAC(49 5∶49 5∶1). The flow rate was 1 0 mL/min. UV detection was performed at 250 nm. The linear ranges of ganoderic acid A(Ⅰ),ganoderic acid C(Ⅱ),ganoderic acid D(Ⅲ) and ganoderic acid E(Ⅳ) were 0 098 3 138,0 091 2 913, 0 089 2 850 and 0 089 2 850 μg respectively. The recoveries of Ⅰ Ⅳ were between 90 4% and 96 83%. The contents of ganoderic acid in culture Ganoderma lucidum and wild Ganoderma lucidum were 0 799 3 555 and 1 619 2 360 mg/g respectively. The present method of PR HPLC is available for determination of content of ganoderic acid in Ganoderma lucidum . The method can be used for the quality control of Ganoderma lucidum preparations for clinical uses. The content of ganoderic acid was not higher in wilding than in culture.
