摘要
目的 :检测软骨细胞培养上清液中是否存在抑制血管生成的因子。方法 :用酶消化、分离鸡胚关节软骨细胞 ,体外单层培养 ,收集细胞培养上清液。并将该上清液作用于小鼠种植性实体瘤 S1 80 及培养人脐静脉内皮细胞 (HU VEC)上 ,探讨其抗血管形成作用的机制。结果 :软骨细胞培养上清液可抑制内皮细胞由 G1 期向 S期转换 ,使细胞停留于 G0 / G1 期。与对照组相比 ,增殖指数 (PI)明显下降 ,瘤重、肿瘤微血管密度 (intratumoral microvessel density,i MVD)也显著降低。结论
Objective:To detemine whether inhibitor of angiogenesis present or not in chondrocytes culture stromal supernatant.Methods:Chick embryo joint chondrocytes digested and isolated by enzyme were grown monolayer culture in vitro.Chondrocyte culture stromal supernatant was collected.The anti angiogenesis mechanisms of the cullture stromal supernatant was studied by the implanted sarcoma S 180 experiment and culture endothelial cell.Results:HUVEC transform G 1 phase to S phase can be decreased by Chondrocyte culture stromal supernatant.Contrast with the control groups,the proliferation indices (PI) decreased evidently,the weight of tumor and the intratumoral microvessel density (iMVD) decrase either.Conclusion:Chondrocytes culture stromal supernatant are the angiogenesis inhibitors.
