摘要
选取王百合种子作为实验材料 ,采用组培法进行繁殖实验。结果表明 ,在相同的温度、湿度和光照条件下 ,应选择颗粒饱满、种皮完好、胚清晰、已过休眠期的成熟种子作为培养材料。接种前种子的消毒时间以 3min为宜 ,适宜的培养基为 1/ 2MS ,附加蔗糖 30g/L、琼脂 6g/L ,pH值为 5 .8~6.2 ,诱导率为 76.7%。最佳继代培养基为MS +NAA0 .0 1mg/L +BA0 .2mg/L。该培养基用于继代培养 ,易形成愈伤组织 ,产生丛生苗 ,繁殖系数每 4 0d为 7.0株。
By tissue culture method with Lilium regale seed,the result shows:under the same conditions of temperature,humidity and illumination,the experimental seed should be plump,with integrated seed capsule and clear embryo,mature and after dormancy.Disinfect the seed 3 minutes before inoculation.The optimum culture medium was 1/2 MS and supplemented with sugar 30g/L and agar 6g/L,which pH was 5.8~6.2 and induce ratio was 76.7%.The best subculture medium was MS and suppplemented with NAA 0.01mg/L and BA 0.2mg/L.Using this kind of subculture medium,it is easy to form callus and produce cluster seedling,the propagation coefficient was 7.0 plants every 40 days.The rootage culture medium of MS was proved good.
