摘要
目的 通过观察雌激素对骨髓来源的成骨细胞所产生的细胞因子的调节作用 ,探讨雌激素抑制破骨细胞功能的机制。方法 在诱导小鼠骨髓细胞分化为成骨细胞后 ,于成骨细胞培养中加入雌激素 ,应用抗体荧光免疫标记法检测成骨细胞培养液中IL 1,IL 6和TNF α水平。结果与对照组比较 ,经 0 0 1~ 1nM雌激素处理后 ,成骨细胞所产生的IL 1和IL 6水平呈浓度依赖性下降 ,TNF α水平则无明显变化。结论 雌激素具有调节成骨细胞分泌细胞因子功能 。
Objective\ To study the mechanism of inhibitory effect of estrogen on osteoclasts through the cell factors produced by osteoblasts. Methods\ The levels of IL 1,IL 6,and TNF α were assessed by immunoassays in mouse marrow derived osteoblast cultures. Results\ In the cultures,the production of IL 1,IL 6,but not of TNF α,was reduced by 0 1 nM estrogen,compared with the controls. Conclusion\ Estrogen is able to modulate the production of several factors by osteoblasts,indicating that estrogen can exert its negative regulation on osteoclast via paracrine mediator(s) produced by osteoblasts.
