摘要 目的 :观察奥氮平对大鼠和小鼠的循环、呼吸和神经系统的药理作用。方法 :大鼠和小鼠均分为 4组 :2 .5 ,5和 10mg·kg-13个剂量组及空白对照组。结果 :奥氮平一次性ig给药可明显降低大鼠的收缩压、舒张压和平均动脉压 ,并可减慢心率 ;明显增加大鼠的呼吸频率 ,降低呼吸幅度 ;明显减少小鼠的自发活动次数 ,增加爬杆的级数 ,且均有较好的量效关系 ;连续给药 1周时 ,也可使小鼠自发活动明显减少 ,大剂量组的爬杆级数明显增加 ,均显示一定的量 效关系。结论 :奥氮平在有效剂量范围内对大鼠的循环系统和呼吸系统均有一定的影响 ,对小鼠的神经系统有明显的抑制作用。 Objectives:To observe the general pharmacology effects of olanzapine in rats and mice.Methods:Animals were divided into four groups:2.5,5 and 10 mg·kg -1 and saline control groups.Results:The systolic,distolic,and mean blood pressures were decreased significantly and the heart rates were lowered at doses of 2.5,5 and 10mg·kg -1 ig once in rats.The frequencies of respiration were increased and the amplitudes of respiration were decreased.Dose related decreases in the times of spontaneous locomotion and increases in the classes of climbing stick were present at the same doses in mice.Similar results were present when ig administered qd for 1 week.Conclusion:Effective doses of olanzapine had effects on circulatory and respiratory systems in rats and inhibitory effects on nervous system in mice.
