平菇子实体分化发育相关基因片段的克隆 被引量:7

摘要 采用RAP PCR差异显示技术对平菇菌丝体及原基、菇蕾和成熟子实体进行研究 ,获得了在原基阶段差异表达的 3条cDNA片段POD1、POD2与POD3 ,经过回收、重扩增、克隆与鉴定后进行测序分析 ,初步的结果表明 3条差异片段的长度分别为 745bp、482bp与 477bp。同时 。 RAP PCR differential display technique was employed to isolate differentially expressed cDNA fragments from mycelium, primodium, young and mature fruit body in Pleurotus ostreatus . Three differentially expressed fragments POD1,POD2,POD3 in primordium were obtained. Their nucleotide sequences were determined and analyzed by Internet BLASTX+BEAUTY searching after these fragments were eluted, reamplified , cloned and identified. Preliminary results showed that the lengths of POD1,POD2,POD3 were 745 bp,482 bp,477 bp respectively. Meanwhile, POD3 was the HSP70 gene fragment.
