摘要
目的 常用的银屑病动物模型主要有两种方法 :即在动物皮肤上诱发银屑病样增生和将银屑病患者皮损移植到裸鼠。本实验旨在通过动物模型验证药物作用机理。方法 利用小鼠鼠尾鳞片表皮的实验动物模型及芳维A酸乙酯的透皮渗透吸收性研究芳维A酸乙酯治疗银屑病的作用。结果 芳维A酸乙酯能显著促进鼠尾表皮颗粒层形成 ,与溶剂对照 (DMSO)和全反式维A酸相比在统计学上有显著性差异 ,且副作用小。结论 芳维A酸乙酯是治疗银屑病的有效药物。
Objective Research of psoriasis has been hampered by the lack of an ideal animal model. The mouse tail model has been used for screening drugs for the treatment of psoriasis.Methods We used this model to evaluate anti parakeratotic activity of arotinoid ethylester.Results The skin was examined histologically for signs of induced granular layers in previously parakeratotic areas,epidermal thicknesses were also measured. Topical arotinoid ethylester could induced remarkedly a granular layer in the tail scales and was more potent than retinoic acid and vehicle solution alone.Conclusion These results show that arotinoid ethylester is an effective drug for treatment of psoriasis.
