12个水稻光温敏核不育系的ISSR标记鉴定及遗传分析 被引量:22

摘要 应用ISSR技术对 12个水稻光温敏核不育系进行DNA指纹分析 ,筛选到 13个多态性丰富的引物 ,共获得 16 9个多态性DNA片段 ,选择其中的 3个引物扩增出的 8个多态性DNA片段可作为鉴定标记 ,能够正确区分供试的 12个水稻光温敏核不育系。根据多态性片段的有无 ,将鉴定标记转换为数字 1和 0 ,建立了 12个水稻光温敏核不育系相应的ISSR标记鉴定识别码。由Nei’s遗传距离创建的聚类分析表明 ,12个材料被聚为两大类群 ,4个粳型不育系聚为一类 ,其余 8个籼型不育系聚为另一类 ,在籼稻群内 ,3个来源于安农S - 1的不育系单独聚为一类 ,与来源于农垦 5 8S的材料有明显的遗传差异。研究结果表明 ,ISSR标记具有简便、快速、多态性丰富等优点 ,可用于构建DNA指纹图谱 ,进行品种鉴定和遗传分析。 In this study, DNA fingerprints of 12 elite photoperiod-sensitive and thermo-sensitive genic male sterile (PGMS and TGMS) rices were analyzed by using inter-simple sequence repeats(ISSR)markers A set of 100 ISSR primers was employed, of which 13 primers showed abundant polymorphisms These 13 primers produced 169 polymorphic bands,and 8 polymorphic bands produced by 3 primers were selected to be used as identification markers, which can help to distinguish these 12 sterile lines. The identification markers were converted into 1 and 0 based on the presence or absence of the polymorphic bands, and ISSR identification code of these 12 sterile lines was constructed. The results of clustering analysis based on Nei's genetic distance revealed that these 12 sterile lines were divided into 2 main groups, 4 japonica sterile lines were clustered into one group and 8 indica sterile lines were clustered into the other group. In the indica group, 3 sterile lines originated from Annong S-1 were clustered together, which showed obvious genetic diversity to sterile lines originated from Nongken 58S. The results showed that ISSR marker was characterized by its simplicity, rapidness and abundant polymorphisms, and suitable for use in fingerprinting construction, identification and genetic analysis.
