FK_(506)缓释膜片促进外周神经再生的实验研究 被引量:14

摘要 目的 观察神经断伤吻合口局部植入FK50 6 缓释膜片对神经再生的影响。 方法 取Wistar大鼠 60只 ,随机平均分为A、B、C、D 4组。切断大鼠右侧坐骨神经后间断吻合 ,将免疫抑制剂FK50 6 溶入聚乳酸 (PLA)后制成 15mm× 8mm× 1mm大小的膜片 ,植入神经吻合口周围作为生物降解缓释制剂 ,可稳定、持续释放FK50 6 约 3周。A组为对照组 ,膜片内不含FK50 6 ;B、C、D组设计释放率分别为1mg·kg 1·d 1、2mg·kg 1·d 1和 5mg·kg 1·d 1。术后第 3、6、10周观察神经吻合口质量、测定坐骨神经功能指数以及电生理学和组织学检查的定性、定量分析。 结果 B、C组测定结果明显优于A组 ,C组优于B组。D组神经连接及再生不良。 结论 适量FK50 6 神经吻合口植入 ,缓释应用对周围神经再生的速度和质量均有明显的促进作用。 Objective To determine the effect of peripheral nerve regeneration through implanting slow releasing diaphragm with FK 506 around the nerve anastomosis Method There were 60 male adult Wistar rats and were divide into A, B, C, D groups randomly by means The right sciatic neve of the animals were cutted and repaired with anastomosis PLA (Polyplactic acid; 15 mm×8 mm×1 mm) with the immunosuppressant FK 506 could be biolysised with stability and persistance in the body for 3 weeks The group A was controlled and there was no FK 506 in it The releasing rates of Fk 506 were 1 mg·kg 1 ·d 1 , 2 mg·kg 1 ·d 1 ,5 mg·kg 1 ·d 1 in B, C, D groups At 3 ,6, 10th weeks post operation, the following items of nerve anastomosis quality, sciatic functional index, electrophisiology and histology were determined with quatitative and quantitative analysis Result The effect of items at B, C group was excellent than that at controlled group significantly, and it was more excellent at group C than at group B The nerve connection at anastomosis and the regeneration were in poor condition at group D Conclusion Application of slow releasing diaphragm which was implanted around the nerve anastomosis with adequate FK 506 , can promoting peripheral nerve regeneration at velocity and quality significantly
