摘要
目的 :探讨芩夏止哮颗粒对卵蛋白 (OVA)致敏哮喘小鼠T细胞活化和细胞因子水平的影响。方法 :用卵蛋白致敏和喷雾攻击复制小鼠过敏性气道炎症模型 ,观察芩夏止哮颗粒对脾细胞增殖反应和mIL 2R表达 ,以及肺泡灌洗液 (BALF)和脾细胞培养上清中IL 4、IL 5和IFN γ水平的影响。结果 :正常小鼠脾细胞自发增殖反应 ,在ConA或OVA刺激下增殖反应 ,以及脾细胞mIL 2R表达均较模型对照小鼠低 (P <0 .0 1)。芩夏止哮颗粒能明显抑制哮喘小鼠脾细胞自发增殖反应和在ConA及OVA刺激下增殖反应及IL 2R的表达 (P <0 .0 1)。模型对照小鼠肺泡灌洗液 (BALF)和脾细胞培养上清中IL 4、IL 5含量显著升高 (P <0 .0 1) ,而IFN γ模型对照脾细胞培养上清中水平低于正常小鼠。芩夏止哮颗粒则能显著降低BALF中和脾细胞分泌IL 4、IL 5的水平 ,提高IFN γ产生水平 (P <0 .0 1或P <0 .0 5 )。结论 :芩夏止哮颗粒治疗哮喘机制与其能抑制哮喘小鼠体内T淋巴细胞的活化 。
To investigate the effects of Qinxiazhixiao Granule (QG) on lymphocyte function and secretion of cytokines in mice model of asthma. Methods: 30 mouse were divided into three groups: the groups of normal control, asthma, QG 8g/kg. The mice model of asthmatic airway inflammation was established by first sensitizing and then challenging the animals with ovalbumin. Proliferation and mIL 2R expression of spleen lymphocytes were observed. Production of IL 4, IL 5 and IFN γ in BALF and supernatant of spleen lymphocyte were detected. Results: Spontaneous proliferation of spleen cells, proliferation with Con A or OVA stimulation as well as mIL 2R expression in the model group were significantly higher than those in the normal group (P<0.01). At the same time proliferation of the spleen lymphocytes in asthma mouse were inhibited and the mIL 2R expression was reduced by QG (P<0.01). Levels of IL 4 and IL 5 in BALF and supernatant of cultured spleen lymphocytes in the model group were markedly higher than those in the normal group (P<0.01). But IFN γ showed production of asthma mouse was lower than that of normal mouse (P<0.01). Compared with those in the model group, IL 4 and IL 5 levels were significantly decreased (P<0.05 or P<0.01) in QG treated mouse. IFN γ level was obviously increased (P<0.01). Conclusions: It showed that these effects of QG might play a key role in treating bronchial asthma. QG can partly prevent asthma by inhibiting T cell activation and regulate the proportion of Th1/Th2 cytokines in the body.
