摘要
目的研制阿霉素异体脱钙骨基质骨粒骨水泥缓释体,并研究分析其体外药物缓释性能及对OS-9001骨肉瘤细胞的抑制能力。方法按Urist等的方法制备异体脱钙骨基质骨粒,通过冻干、真空吸附等处理,将阿霉素载入其中并与骨水泥按1:1比例复合,制得阿霉素异体脱钙骨基质骨粒骨水泥缓释体,对该缓释体进行体外药物释放及其浸出液的体外抑瘤试验。结果该缓释体第1天释放量为总含量的19.23%,其第1、20、40及70天的浸出液对OS-9901骨肉瘤细胞的抑制率分别为64.27%、41.68%、28.71%及24.32%。结论该缓释体具有良好的缓释功能,在70d内对OS-9901骨肉瘤细胞维持良好有效的抑制率。
Objective To prepare decaleified bone matrix particles(DBMP) with adriamycin impregnated bone cement(BC) drug delivery system(DDS).It drug delivery characteristic and inhibitory ability on human osteogenic sarcoma cell OS 9901 were studied and analysed in vitro.Methods DBMP were made according to the methods of urist et al.Adriamycin was putting into DBMP by vacuum absorption and feeaze drying techniques.Then the DBMP with adriamycin were impregnated BC mass same ratio and formed DDS.The release test of DDS and exudant in inhibiting human osteogenic sarcoma cell OS 9901 were examined in vitro.Results The DDS had an explosive releasing within the first 24 hours,account for 19.23%.The inhibitory ratio of the first,20th,40th and 70th day was 64.27%,41.68%,28.71% and 24.32% respectively in vitro.Conclusions The DDS had good sustained release function.It keeps excellent the inhibitory ratio on human osteogenic sarcoma cell OS 9001 in 70 days in vitro.
