生长抑素对人胰腺癌细胞株HS766T的生长调控 被引量:1

摘要 目的观察生长抑素对人胰腺癌细胞株HS76 6T的生长抑制作用并探讨其作用机制。方法细胞培养 ,分别加入不同浓度的生长抑素 ,应用四甲基偶氮唑盐法观察细胞增殖程度 ;放射免疫法测定细胞内cAMP含量 ;Fura 2 AM测定细胞内Ca2 +浓度。结果不同浓度的生长抑素 (10 - 6~10 - 1 2 M)均能有效地抑制HS76 6T的生长 ,能抑制HS76 6T细胞内cAMP生成和Ca2 +水平 ,分别在 5min时和 3min时作用最大 ,cAMP生成量和Ca2 +水平与生长抑素浓度呈负相关。它能使垂体腺苷酸环化酶激活多肽 (PACAP)诱导的细胞内cAMP生成量和Ca2 +减少。结论生长抑素能抑制人胰腺癌HS76 6T细胞的生长 。 ObjectiveTo observe the growth inhibition of somatostatin (SS) on human pancreas carcinoma cell strains HS766T and its mechanism.MethodsHS766T cells cultured in DMEM medium containing 10% FCS were treated with SS.Proliferation and viability of cell was measured by the Mosmann′s method (MTT). Intracellular adenosine monophosphate levels was detected by radioimmunoassay. Intracellular Ca 2+ level were measured using a F-2000 flurorescence spectrophotometer.ResultsSS inhibited the proliferation of HS766T in a concentration dependent manner (P<0.01,r=-0.921). SS inhibited production of intracellular cAMP and Ca 2+ in HS766T cells. The production was negatively correlated with the concentration of SS (P<0.01). SS inhibited cell proliferation, intracellular level of cAMP and Ca 2+.ConclusionsThe proliferation of HS766T was inhibited by SS. SS also suppresses PACAP 1-38 induced cell proliferation.
