榄香烯及榄香烯复合瘤苗对小鼠腹腔巨噬细胞功能影响的分析 被引量:1

摘要 目的 :分析小鼠腹腔巨噬细胞 (peritonealexudativemacrophage,PEMφ)在榄香烯复合瘤苗免疫保护效应中的作用。方法 :用血红蛋白释放法、硝酸还原法、MTT法等检测榄香烯及榄香烯复合瘤苗对小鼠腹腔巨噬细胞的吞噬、一氧化氮的产生、杀瘤以及对脂多糖 (lipopolysaccharide,LPS)和粗提淋巴因子刺激的反应等功能的影响。结果 :低浓度 (5 ,0 .5 μg/ml)的榄香烯和榄香烯复合瘤苗在体外可以增强PEMφ的活性 ,但瘤苗及其HSP70 -肿瘤肽免疫后其PEMφ的上述功能未见变化。结论 :榄香烯复合瘤苗的免疫保护效应并非通过PEMφ而实现。而短小棒状杆菌作为榄香烯复合瘤苗的佐剂 。 Objective: To analyse the function of the peritoneal exudative macroghages ( PEMφ) in the immunoprotective effect of elemene combo-TCV. Methods: we examined the effect of elemene and elemene combo-TCV on the functions of PEMφ including their cytotoxcity against tumor target cells by MTT assay, nitric oxide(NO)production by NO assay kit, phagocytosis activity by hemoglobin enzyme release assay (Hb-ERA) and their response to lipopolysaccharide(LPS)or crude lymphokine(LK)stimulation. Results: Low concentration(5, 0.5 μg/ml)elemene and elemene combo-TCV could in vitro enhance the activity of PEMφ. But the functions of PEMφ obtained from TCVs and their HSP70-tumor peptide complexes immunized mice did not show significant change as compared with those of PEMφ from non-immunized mice. Conclucions: The active immunoprotective effect of the elemene combo-TCV was not achieved through PEMφ, while CP, as an adjuvant of elemene combo-TCV, could obviously increase the number and enhance the functions of PEMφ.
