NP-470和DDP联用对小鼠腹水瘤腹水生成的抑制作用

摘要 目的 :为了验证 0 -(氯乙酰 -氨甲酰基 )烟曲霉醇 (TNP-470 )和顺铂 (DDP)联用的增效作用。方法 :将 64只 61 5小鼠腹腔内接种腹水瘤细胞株后 ,随机分为 4组 ,然后腹腔内分别注射生理盐水(第 1组 ) ,DDP(第 2组 ) ,TNP -470 (第 3组 )及联合用药 (第 4组 ) ,观察瘤细胞株对各组小鼠的腹水生成 ,瘤细胞数 ,存活率及生存时间。结果 :DDP,TNP -470及联合用药组与生理盐水对照组相比可显著抑制 61 5小鼠腹水的生成 ,减少瘤细胞数量 ,延长生存期 (P <0 .0 5 ) ;联合用药组与其它 3组比较在瘤细胞计数和瘤细胞存活率方面均有显著性差异 (P <0 .0 5 ) ;在小鼠存活率及生存时间方面 ,联合用药组延长最明显 ;在用药过程中 ,联合用药组可引起体重下降 ,但停止用药后体重又可逐渐得到恢复。结论 :TNP-470 ,DDP对小鼠腹水瘤腹腔种植后腹水的生成都有明显的抑制作用 ;TNP -470和DDP联用能起增效作用 ; Objective: To determine whether O-(chloroacetyl-carbamoyl)fumagillol (TNP-470) could potentiate the anti-tumor effect of cisplatin (DDP). Methods: Sixty-four inbred 615 mice were inoculated with ascitogenous tumor cells of HCa-P lines and were divided into 4 groups at random: Group 1, normal saline; Group 2, DDP; Group 3, TNP-470; Group 4, combination. Production of ascites, number of tumor cells, survival rate of tumor cells and survival period of mice were evaluated. Result: DDP, TNP-470 and combination obviously inhibited the production of ascites and reduced the number of tumor cells and prolonged the survival period of tumor-loading mice(P<0.05).The number of tumor cells and survival rate of tumor cells of the combination group were significantly different from those of the other 3 groups(P<0.05).The combination group was the most outstanding on the survival rate and the survival period of tumor-loading mice. During the period of administration, the combination group showed weight loss, without abnormal activity. After suspending administration, the weight could regain gradually. Conclusion: These results suggested that the combination of TNP-470 and DDP might produce additive effect. Although leading to weight loss, this was reversible.
