摘要
以健壮燕麦 (Avera sativa L .)为供体 ,宁春 4号小麦为受体 ,采用花粉管通道法进行外源 DNA导入。对供体、受体及其 D5代的变异株系进行酯酶同工酶和过氧化物酶同工酶分析表明 :变异后代与亲本的谱带有明显差异 ,无论是在酯酶同工酶中还是在过氧化物酶同工酶中 ,均出现了供体特有的谱带 ;在过氧化物酶同工酶中既出现了供、受体均不具有的新的杂交酶带 ,有的还显现了偏父或偏母以及完全互补型的酶带 ;酶的活性也发生了很大变化。认为可以用同工酶分析作为鉴定外源 DNA导入的生化指标之一。
Total DNA introduction was studied using pollen tube passage way with oat (Avena sativa L.) as donor and wheat as recipient. The asterase isoenzyme and peroxidase isoenzyme patterns of variant strains in D\-5, donor and receptor were analyzed. The enzyme bands between donor, recipient and their progenies showed obvious difference. There are special bands of donor in the enzyme pattrns of progenies of both esterase isoenzyme and peroxidase isoenzyme. Some new hybrid enzyme bands as well as tendency towards donor or recipient or complete complementary enzyme bands appeared in the peroxidase isoenzyme and the activities of enzyme also took place great change. Therefor, the authors considered that the isoenzyme can be used as one of the biochemical indicator for evaluating wheat having received oat DNA.\;
