介绍一种改进的高效液相色谱柱的装填方法——稀匀浆装填方法 .该方法使用无毒、低粘度的稀匀浆溶液和较长的合适结构的匀浆罐 .在高于 70 MPa的压力和 5 0 m l/min匀浆输送流量下 ,装填 5 μm ODS填料的反相色谱柱和 Si O2 硅胶的正相色谱柱都获得良好的结果 .柱效可达到 60 0 0 0 /m以上 ;峰对称性好 ,SI=1~ 1 .2 5 ;寿命长 ,经过 5 0
The improved packing method of columns of high performance liquid chromatograp hy, or the diluted homogenate slurry packing method is introduced. A non toxic slurry solvent with low viscosity, and a longer shape slurry reservoir are used. The excellent performance HPLC columns packed with 5μm ODS bonded stationary phases in reverse phase or SiO 2 silica gel stationary phases in right phase are obtained. The columns' performance is good, with a high N ≥60000/m, the symmetry of chromatographic peaks is excellent SI=1~1.25; the life is longer, after analysis of 500 samples, the columns' performance and the symmetry of peaks are steady. This packing method is fully reproducible and efficient to get high performance HPLC columns.
