V_C、V_E对家兔辐射致血液几种免疫细胞损伤的影响及剂量效应关系 被引量:2

摘要 目的  探讨 60Co 辐射家兔造成外周血免疫系统损伤后维生素C(Vc)和维生素E(VE)对外周血中淋巴细胞、粒细胞、单核细胞的影响及剂量效应关系。方法以健康家兔随机分为11组,每组6只,分别为:阴性组、阳性组、Vc实验组和VE实验组。阴性组与阳性组分别灌胃花生油,静脉注射生理盐水,Vc通过静脉注射给药,VE 为灌胃法给药。Vc的剂量组为:10 mg·kg-1,20 mg·kg-1,Vc 40   mg·kg-1,Vc 80 mg·kg-1;VE 的剂量组为:5 mg·kg-1,10 mg·kg-1,20 mg·kg-1,40 mg·kg-1,80 mg·kg-1。Vc,VE 连续给药5 d。于试验第3 d阳性组及各Vc、VE给药组的家兔进行4.5 Gry的60Co照射,试验第5 d家兔麻醉后剖腹取腹腔静脉血,分别测定外周血几种免疫细胞数量变化。结果未经任何保护阳性组的淋巴细胞(LYM)高于阴性组的百分数量(P<0.05),Vc和VE给药各组的LYM均低于阳性组。经辐射作用后LYM数量随Vc剂量增加呈现轻微下降趋势,单核细胞(MON)数量无明显变化,而粒细胞(GRA)数量随Vc剂量增加呈现一定的上升趋势后,? Aim To study the effects of vitamin C(Vc) and vitamin E(VE) on immune hemacytes lymphocyte(LYM),granulocyte(GRA), monocyte(MON) of rabbits injured by radiation of 60Co and their relationship between dose-effect. Method Chinese rabbits were randomly divided into eleven groups,each group contained six rabbits. They were negative control group, positive group, trial groups of Vc, VEgroups containing four different dosage of Vc and five different dosage of VE . The four vitamin C groups were administrated by injection of vein with the dosages of Vc 10 mg·kg-1, Vc 20 mg·kg-1, Vc 40 mg·kg-1, Vc 80 mg·kg-1 everyday respectively. The five vitamin E groups were administrated by gavages with the dosage of VE 5 mg·kg-1, VE 10 mg·kg-1, VE 20 mg·kg-1, VE40 mg·kg-1, VE 80 mg·kg-1, everyday respectively too. On the third day all groups except negative control group were radiated by 60 Co with the dose of 4.5 Gry. On the sixth day all rabbits were killed and blood was taken for determination of LYM, GRA and MON percentage and number changes. Results After radiation, the percentage of LYM in positive group was higher than the negative group(P<0.05), and the percentage of Vc groups and VEwas lower than that in positive group. The LYM number of the hemacytes decreased gradually with the of the increase of Vc dosage. No significant difference was observed in numbers of the MON. The proportion of GRA was higher than that of positive group with the rise of the Vc dosage in certain range. The proportion and the number of LYM on were reduced with the increase of VEdosage. But there was no significant change of MON too. The proportion and number of GRA was also rising but then decreased with the rise of the dosage of VE. ConclusionWhen Vc or VEware isolate administrated to rabbits injured by radiation of 60Co, there was a little protective effect on immune hemacytes LYM,GRA and MON. The concept of "Multi-antioxidants chai"or "Antioxidants network"in study of the protective effect of antioxidant against injury of radiation was impotent.
