摘要
目的:研究内毒素血症中诱导型一氧化氮合酶(iNOS)对大鼠肠系膜淋巴管微循环动力学的影响。方法:给大鼠注入脂多糖(LPS)和S-甲基异硫脲(SMT),用倒置显微镜和图像处理系统观察及测量大鼠淋巴管的口径和运动频率,用免疫组化染色法观察iNOS在大鼠淋巴管的表达。结果:LPS组肠系膜淋巴管口径比正常扩大,运动频率降低,iNOS阳性细胞增多。而LPS+SMT组的淋巴管口径和运动频率未见明显变化,iNOS阳性细胞较LPS组显著降低。结论:iNOS过量表达影响了淋巴循环;SMT可通过抑制iNOS的活性而对内毒素血症期间的淋巴管起保护作用。
Objective:To study the effects of inducible nitri c oxide synthase(iNOS)on the lymphatic microcirculation in endotoxemia. Methods:Rats were injected with lipopolysaccharides(LPS) and SMT .The diameters and motive frequency of the rat mesenteric lymphatic vessels wer e observed and measured by inverted microscopy and digital image processing and i NOS was assessed by immunohistochemistry staining.Results:The diamet ers of lymphatic vessels dilated and frequency decreased compared with those bef ore injection in LPS group. LPS treatment strongly induced iNOS in epithelial ce lls of mesenteric lymphatic vessels. There were no obvious changes among diamete rs and frequency in LPS+SMT group.SMT caused a reduction of iNOS expression and NO production. Conclusion:iNOS may have an effect on regulating the motion of lymph atic vessels. SMT may have a protective effect by reducing the activity of iNOS during endotoxemia.
