雷公藤提取物对增生性瘢痕成纤维细胞的负性调节作用

NEGATIVE REGULATION OF TRIPTERYGIUM WILFORDII HOOK ON HYPERTROPHIC SCAR-DERIVED FIBROBLASTS

目的 :观察雷公藤提取物对成纤维细胞形态和增殖的影响。方法 :从增生性瘢痕中获取的成纤维细胞进行体外培养 ,加入 5× 10 - 3mg/ ml的雷公藤提取物作用 2 4 h后 ,在光镜下观察细胞形态。并用 MTT法检测成纤维细胞与不同浓度的雷公藤提取物 ( 5× 10 - 3、5× 10 - 4、5× 10 - 5、5× 10 - 6 mg/ m l)作用 2 4 h后增殖活性。结果 :雷公藤提取物在对成纤维细胞无毒性作用的浓度下 ,可使成纤维细胞的突起变短 ,增殖减缓。结论

Objective: The experiment is used to observe the effect of TWH on the morphology and the proliferation of fibroblasts. Methods: Fibroblasts obtained from hypertrophic scar were cultured in vitro. Morphological changes were observed under optic microscope after fibroblasts were preincubated with TWH(5×10 -3 mg/ml)for 24h. And the proliferation of the fibroblasts was examined by MTT method after fibroblasts were preincubated with TWH(5×10 -3 ?5×10 -4 ?5×10 -5 ?5×10 -6 mg/ml)for 24h.Results: The number of fibroblasts becomes less and the processes of fibroblasts become shorter after TWH action. The proliferation of the fibroblasts was inhibited. This inhibition of TWH on fibroblasts was proved that it was not toxicity of this drug. Conclusion: TWH can inhibit the fibroblasts not only on morphology but also on proliferation.
