摘要 目的 初步研究血管壁脂质过氧化状态下FN、P10 5基因表达的变化及其意义。方法 以不同剂量的盐阿霉素 (adriamycin ,ADR)分别一次性腹腔注射雄性SD大鼠 ,制备脂质过氧化模型。采用硫代巴比妥酸 (TBA)荧光分光光度法测定大鼠血清脂质过氧化产物丙二醛 (malondialdehyde ,MDA)含量。运用逆转录聚合酶链反应 (RT PCR)测定大鼠主动脉中的Fibronectin(FN)、P10 5coactivatormRNA的表达。结果 ADR中、高剂量组大鼠血清中MDA含量高于对照组 (P <0 .0 5 ,P <0 .0 1) ;RT PCR结果表明FN、P10 5mRNA在不同剂量模型组呈不同程度的高表达。结论 在阿霉素导致血管壁脂质过氧化后 ,有FN、P10 5基因的高表达 。 Objective To study the changes of PN and P105 expressions in arterial wall injury induced by lipidperoxidation.Methods The rat model of arterial wall injury induced by lipidperoxidation was prepared by intraperitoneal injections of Adriamycin in various dosages.The serum content of malondialdehyde (MDA) was estimated with thiobarbituric acid.```````` .RT?PCR was performed to analyze the expressions of fibronectin mRNA(FN) and P105 coactivator mRNA in the aorta.Results MDA contents in the middle and high ADR dose groups were higher than that in the control group ( P <0.05, P <0.01).FN mRNA and P105 mRNA expressions were higher in all ADR groups.Conclusion The higher expressions of FN and P105 are shown in all rat models of arterial wall injury induce by intraperitoneal injections of Adriamycin.It is proposed that lipidperoxidation might induce arteriosclerosis through multiple pathways.
