摘要
目的 :探讨尼莫地平与甘露醇联合治疗局灶性脑缺血再灌注大鼠的神经保护作用。方法 :采用线栓法制备大鼠大脑中动脉缺血 2h再灌注 2 4h模型并术前 15min分别静脉注射生理盐水、2 0 %甘露醇、尼莫地平以及 2 0 %甘露醇和尼莫地平 ,通过TTC、TUNEL染色分别观察梗死灶体积和凋亡细胞数目的改变。结果 :甘露醇与尼莫地平联合治疗组梗死灶体积显著缩小、凋亡细胞数目显著降低 (P <0 .0 5 )。结论 :尼莫地平与甘露醇联合治疗可通过抗细胞凋亡机制发挥对局灶性脑缺血再灌注大鼠的神经保护作用。
Objective: To evaluate the neuroprotective effects of nimodipine combined with mannitol in the treatment of transient focal cerebral ischemia/reperfusion. Methods: The rat model of middle cerebral artery occlusion (MCAO) for 2 h and reperfusion for 24 h was established by using thread embolism. The rats in the group Ⅰ was treated with 0.9% saline, those in the group Ⅱ received 20% mannitol, those in the group Ⅲ received nimodipine intravenously 15 min before MCAO and those in the group Ⅳ received both 20% mannitol and nimodipine intravenously. At the end of experiments, the brain was taken and sectioned in coronal slices. The slices were stained with TTC and with the transferase dUTP nick end labeling (TUNEL) technique, respectively. Results: The infarct size in the groups Ⅱ, Ⅲ and Ⅳ was significantly decreased as compared with that in the group Ⅰ ( P < 0.01 ). The number of apoptotic cells was statistically lower in the group Ⅲ than other three groups ( P < 0.05 ). Conclusion: Combined use of mannitol and nimodipine provides neuroprotection by preventing both the necrotic and apoptotic components of cell death after transient focal cerebral ischemia/reperfusion.
