转化生长因子β_1和骨形成蛋白2体外诱导成牙本质细胞分化 被引量:4

摘要 目的 观察转化生长因子 β1(transforminggrowthfactorβ1,TGF β1)和骨形成蛋白 2 (bonemorphogeneticprotein 2 ,BMP2 )对体外培养的鼠牙乳头成牙本质细胞分化的影响。方法 取 17d胎龄小鼠下颌第一磨牙牙胚 ,胰蛋白酶消化分离牙乳头 ,置半固态培养基培养 6d ,半固态培养基中加入重组TGF β1或BMP2与肝素 ,组织学观察。结果 TGF β1或BMP2加肝素可诱导牙乳头周边细胞发生极化 ,并分泌胞外基质。TGF β1或BMP2单独加入时未见细胞极化 ,但基质分泌增加。结论 TGF β1和BMP2均能诱导成牙本质细胞的细胞学分化和分泌功能。 Objective To investigate the effects of transforming growth factor β 1(TGF β 1)and bone morphogenetic protein 2 (BMP2) combined with heparin on odontoblast differentiation. Methods Trypsin isolated dental papillae from day 17 mandibular first molar were cultured in semisolid agar medium for 6d. Recombinant human TGF β 1 and BMP2 combined with heparin were added to the medium. Results TGF β 1 and BMP2 combined with heparin induced differentiation of odontoblasts and promoted matrix secretion. Odontoblast differentiation never occurred when TGF β 1 or BMP2 were added alone to the medium, whereas an increase in extracellular matrix production was observed. Conclusion These results demonstrate that both TGF β 1 and BMP2 stimulate the cytological and functional differentiation of preodontoblasts, and that heparin might play important role as a substrate.
