摘要
目的 :研究淫羊藿、枸杞子对老年大鼠线粒体DNA缺失、线粒体呼吸链酶复合体及ATP合成的影响。方法 :Wistar系雄性大鼠 ,幼年大鼠 (1月龄 )、青年大鼠 (4月龄 )每组均为 1 2只 ;老年大鼠 (2 2月龄 ,31只 ) ,随机分为对照组 (1 0只 )、淫羊藿组 (1 0只 )和枸杞子组 (1 1只 )。用聚合酶链反应 (PCR)、酶动力学和生物发光技术进行检测。结果 :淫羊藿、枸杞子可减少老年大鼠心、脑、骨骼肌组织线粒体DNA缺失 ,提高心、脑线粒体三磷酸腺苷(ATP)的合成和心、骨骼肌线粒体呼吸链复合酶Ⅳ活力及脑线粒体呼吸链复合酶Ⅰ活力 ;淫羊藿还可提高脑线粒体呼吸链复合酶Ⅳ活力和骨骼肌线粒体ATP的合成。结论 :淫羊藿。
Objective: To investigate the effect of Herba Epimedii and Fructus Lyci i on mitochondrial DNA deletion,activity of respiratory chain enzyme complexes and ATP synthesis. Methods: Wistar male rats were used in the study,one month old,four month old and 22 month old.22 month old rats were randomly divided into three groups. Senility group was treated with normal saline, Herba Epimedii group was treated with Herba Epimedii,Fructus Lycii group was treated with Fructus Lyci .Polymerase chain reaction(PCR), enzyme kinetics and bioluminescence techniques were used. Results: In aged rats, treatment of Herba Epimedii(Epimedium) and Fructus Lycii (Wolfberry Fruit) reduced the ratio of deleted / normal mtDNA in brain, skeletal muscle and heart. The treatment increased the rate of ATP synthesis in heart and brain, the activity of mitochondrial respiratory chain enzyme complexes Ⅳ in heart and skeletal muscle, and the activity of mitochondrial respiratory chain enzyme complexesⅠin brain. Treatment of Herba Epimedii alone also increased the activity of mitochondrial respiratory chain enzyme complexes Ⅳ in brain and the rate of muscular ATP synthesis. Couclusion: Treatment of Herba Epimedii and Fructus Lycii protected aged rats from oxidative damage to mitochondria.
