摘要
采用Ca2+指示剂Fura2作为细胞内钙离子的荧光探针,用荧光分光光度计检测了黄芩甙对培养的大鼠主动脉平滑肌细胞内游离钙浓度Cai+2的影响。结果表明,随黄芩甙浓度的增加,可剂量依赖性地显著降低平滑肌细胞静息Cai+2,并显著抑制去甲肾上腺素和高K+引起的细胞Cai+2的升高,提示黄芩甙可通过阻断平滑肌细胞膜上的电压依赖型钙通道和受体操纵型钙通道,抑制细胞Cai+2的增高,这可能与其降压作用机制有关。
The effects of Bai on intracellular free calcium concentration Cai+2 was observed by Fura2 fluorescence measurements in cultured vascular smooth muscle cells (VSMC) of rats.The results indicated that Bai could decrease basal Cai+2 of VSMC concentration dependently,and inhibit the increase of Cai+2 induced by NE and high K+ significantly.It suggested that Bai probably inhibited the increase of Cai+2.by blocking PDC and ROC on the VSMC membrane.That might be related to the mechanism of its lowering blood pressure.
