灵芝多糖对小鼠巨噬细胞内三磷酸肌醇和二酰基甘油的作用研究 被引量:17

摘要 为研究灵芝多糖(GLB7) 对小鼠腹腔巨噬细胞( MΦ) 三磷酸肌醇(IP3) 和二酰基甘油(DAG) 的作用,确定其对MΦ信号转导途径,采用细胞培养、放射免疫分析及离子交换层析和薄层层析法测定了小鼠腹腔MΦ中IP3 和DAG 的变化。结果显示GLB7 能引起小鼠MΦ中IP3 和DAG 浓度升高。IP3 的达峰时间为30s,百日咳毒素(PTX) 对此现象有抑制作用;DAG 的合成出现两个峰,第一个峰迅速短暂,峰值位于30s,第二个峰是持续近2min 的迟发峰。表明IP3/Ca2 + 和DAG/PKC Objective: Through investigation the effect of GLB 7 on inositol trisphosphate (IP 3) and diacylglycerol (DAG) in murine peritoneal macrophages to provide a scientific evidence for it's immnomodulatory mechanism.Method: Cell culture, radioimmunological asssy, anion-exchange columns and thin-layer chromatography were used.Result: GLB 7 increased the production of IP 3 and DAG in murine peritoneal macrophages.The peak time of IP 3 was 30s, pertussis toxin (PTX) could inhibit GLB 7 from increasing the production of IP 3. In the production of DAG, two peak had been seen, the first peak was rapid and transitory, and the second was sluggish.Conclusion: The both path of signal transduction of IP 3/Ca 2+ and DAG/PKC are participated in the immunomodulatory of GLB 7 to macrophages.
