摘要
本研究采用高频刺激大鼠海马脑片Schafercolateral-CA1锥体细胞突触传递以诱导长时程增强效应(LTP),观察樟柳碱对其的影响,结果表明樟柳碱能明显抑制该区LTP的形成,而对单刺激引起的场电位则没有影响,提示樟柳碱对LTP的抑制作用可能是其导致记忆获得不良的细胞机制之一。
Anisodine, a muscarinic receptor blocker with a structure similar to scopolamine, was first isolated by Chinese scientists. It was found that anisodine could induce memory impairment in clinic and therefore was used to make animal memory impaiment model. But the cellular mechanism is unclear.Long term potentiation (LTP) is a prolonged form of activity dependent synaptic plasticity that is thought to be related to learing and memory and widely applied to study the cellular mechanism of learning and memory. In this paper we investigated the effect of anisodine on the LTP generation by the method of tetanization in the schaffer collateral CA1 pyramidal cell. Excitatory field potentials (EPSP) were recorded in living rat hippocampus slice. Anisodine (10 -5 M) showed inhibition to the generation of LTP without influence on the field potential by sigle stimulation. These results indicate that anisodine inhibits LTP may play role in its inducing memory impairment.
