黄芪黄酮对低渗造成的血管内皮细胞单层通透性增高的改善作用 被引量:15

摘要 新生牛主动脉分离获得单个内皮细胞培养于微孔滤膜上直至形成致密单层。通过灌流含 5 g/L白蛋白的Hanks液后 ,测定经低渗溶液处理 (将M1 99溶液中含的 2 0 %血清改成 2 %血清 )或经低渗溶液加 1 0 4g/ml黄芪总黄酮或 1 0 4g/ml黄芪毛蕊异黄酮处理的液体滤过系数 (Kf)、液体滤过流量 (Jv)和蛋白质渗透压反射系数 (σ)。内皮单层经低渗溶液处理后 ,Kf和Jv降低 ,σ升高 ;而总黄酮和毛蕊异黄酮能抑制因低渗造成的Kf和Jv的降低及σ升高。形态学分析表明总黄酮、毛蕊异黄酮能够抑制因低渗造成细胞肿胀和细胞间距离的增宽。结果表明低渗能够引起血管内皮单层通透性的增加 ,而黄芪黄酮能够减轻低渗造成的内皮单层通透性的增加。 We studied effect of flavonids isolated from Astragalus membrananceus on the increase of endothelial permeability induced by low osmolarity. The endothelial cells isolated from bovine aorta were cultured on polycarborate microporous filter membrane to develop confluent endothelial monolayer. Fluid filtration coefficient (Kf), filtration volume (Jv) and osmotic reflective coefficient (σ) to protein were measured in the endothelial monolayer treated by low osmolarity solution (decrease concentration of serum in M199 from 20% to 2%) for 120 min, or treated by low osmolarity solution containing 10~ -4 g/ml total flavonid or calycosin for 120 min after perfused Hanks balanced salt solution containing 5g/L albumin. The findings showed that in the endothelial monolayer treated by low osmolarity solution Kf, Jv were increased and σ decreased after perfused Hanks balanced salt solution containing 5g/L albumin. Application of total flavonid or calycosin in the endothelial monolayer treated low osmolarity solution induced reduce of Kf, and Jv and σ enhance after perfused Hanks balanced salt solution containing 5g/L albumin, compared with in the endothelial monolayer treated low osmolarity solution purely. Morphological analysis demonstrated that flavonid and calycosin can inhibit widening of intercellular distance and enlargement of cellular area in the endothelial monolayer induced by low osmolarity. The results indicated that increases in the endothelial monolayer permeability induced by low osmolarity solution, and total flavonid or calycosin can inhibit increases in the endothelial monolayer permeability induced by low osmolarity solution. It is necessary that the cellular mechanism of permeability response induced by total flavonid or calycosin remain to be further elucidated.
