脑力智宝对兴奋性氨基酸和一氧化氮引起的PC12细胞损伤的保护作用 被引量:5

摘要 目的 :观察脑力智宝对神经细胞损伤的影响。方法 :采用血清药理学方法 ,体外培养PC12细胞 ,研究脑力智宝对兴奋性氨基酸和一氧化氮引起的PC12细胞损伤的影响。结果 :脑力智宝提取物及其含药血清可明显地对抗兴奋性氨基酸和一氧化氮引起的PC12细胞形态改变 ,降低胞外乳酸脱氢酶活性 ,增加细胞存活率。结论 Objective: To investigate the effect of glutamate (Glu) and sodium nitroprusside (SNP) on cultured PC12 cells and the protective effects of NaoLiZhiBao (NLZB). [WT5”HZ]Methods:[WT5”BZ] Serologic pharmacological test was used to study effect of NLZB on Glu induced and SNP induced PC12 cells injury. The viability of PC12 cells was determined by MTT colormetric microassay and the overall cell injury was assessed by measurement of lactate dehydrogenase (LDH) activity. [WT5”HZ]Results:[WT5”BZ] The death rate of PC12 cells was increased and the release of LDH into the bathing medium increased obviously when exposed to Glu and SNP, inducating that the PC12 cells were seriously damaged. When the cells were pretreated with the extract of NLZB and the serum of containing drug, The values of LDH efflux in culture medium and the death rate of PC12 cell were singnificantly decreased. [WT5”HZ]Conclusions:[WT5”BZ] Our finding suggest that NLZB can antagonize the neurotoxicity of excitatory amino acids and NO.
