摘要
目的 观察刀豆蛋白A (ConA)诱导的小鼠急性肝损害肝组织内粘附分子 -1(ICAM -1)的表达 ,以研究ICAM -1在这种肝损害模型中的作用。方法 ConA以 18mg/kg的剂量诱导小鼠急性肝损害 ,用免疫组化的方法观察 8h后肝组织ICAM -1的表达 ,并观察血清ALT的变化。结果 8h后出现明显肝损害 (ALT水平升高 ) ,肝组织内ICAM -1表达明显增强。结论 ICAM -1参与了ConA小鼠肝损害的病理过程。
Objective To investigate the possible involvement of intercellular adhesion 1 molecules(ICAM 1) in the mechanisms of ConA induced acute liver injury in mice.Methods 18 mg/kg of ConcanavalinA was intravenously injected into male KunMing mice in order to induce acute liver faliure.8 houss later serum ALT was measured and the expression of ICAM 1 in the liver was detected by immunohistochemical method.Rosults There were Signs of hepatitis with elevated transaminase activity in plasma,severe infiltration of the liver by granulocytes and monocytes.Necrotic areas in the liver were observed in ConA injucted mice.The expression of ICAM 1 after the injection of ConA was significantly increased not only in hepatic sinusoidal lining cells,but also on the surface of hepatocytes.Conclusion The expression of ICAM 1 is involved in the mechanism of ConA induced acute liver injury in mice. [
