褪黑素对急性实验性脊髓损伤的保护作用 被引量:5

The protective effects of melatonin on acute experimental spinal cord injury

摘要 目的 观测褪黑素对急性实验性脊髓损伤的保护作用。方法 SD大鼠随机分 3组 ,一组椎板切除不损伤脊髓 ,另两组Allen s法中度量 (5 0g/cm)损伤脊髓 ,分别予褪黑素 10 0mg/kg体重腹腔注射和含 5 %乙醇的生理盐水腹腔注射 ,4h后取伤段脊髓 ,测量组织的MDA ,游离铁和谷胱甘酞过氧化物酶活性(GSH PX)。结果 损伤后 4h ,伤段脊髓组织的游离铁有明显的升高 ,导致丙二醛 (MDA)大量生成 ,组织GSH PX活性升高 ;褪黑素能降低脊髓损伤后游离铁的释放 (P <0 0 5 ) ,减少MDA的生成(P <0 0 5 ) ,抑制组织谷胱甘酞过氧化物酶活性升高 (P <0 0 5 )。结论 褪黑素能减轻脊髓损伤后游离铁浓度升高触发的脂质过氧化反应。 Objective To observe the protective effects of melatonin on acute experimental spinal cord injury in rats.Methods Sprayue Dawleys rats were randomly divided into three groups,rats in group one only had laminectomy performed and nontraumatized spinal cord samples obtained;rats in other 2 groups were injured by Allens weight drop method(50g/cm),then were given either a 100mg/kg single does of melatonin or saline containing 5% ethanol intraperitoneally immediately.the levels of free iron,MDA and GSH-PX of involved spinal cord segments 4 hours after the injury were measured.Results In 4 hours after SCI,the level of free iron,MDA and GSH PX increased,melatonin can attenuate the changes(P<0 05).Conclusion Melatonin can reduce the levels of iron induced lipid peroxidationa fter spinal cord injury. [
