司帕沙星对戊巴比妥钠催眠作用的影响

摘要 以戊巴比妥钠 (PBS)催眠时间作为细胞色素 P4 5 0 (CYP)体内活性指标 ,观察司帕沙星 (SPX)对小鼠 CYP的影响。结果 SPX单次给药 (5 0 ,10 0 mg·kg-1,ig) 1h后 ,使 PBS催眠时间显著延长 ,但延长程度小于西米替丁组。SPX连续给药 5 d (5 0 ,10 0 mg· kg-1·d-1,ig) ,在末次给药 36 h后 ,则见 PBS催眠时间缩短 ,但缩短程度小于苯巴比妥组。提示 SPX单次给药后可能抑制 CYP,但连续给药对 CYP有诱导作用。 The test was done to investigate the influence of sparfloxacin (SPX) on mice cytochrome P450(CYP) with hypnogenesis by pentobarbital sodium(PBS). The hypnosis duration of SPX groups was prolonged significantly, one hour after given SPX(50, 100 mg·kg -1 , ig), but less than that of cimetidine group. Howerer, when given SPX(50, 100 mg·kg -1 ·d -1 , ig) for 5 days, the shortened hypnosis duration was observed 36 hours later at the last administration, which is not so obvious as that of phenobarbital. The results suggested that single-administation of SPX could inhibit CYP, but SPX might also induce CYP if it was given repeatedly for several days.
