摘要
通过建立HPLC-ELSD法检查猪去氧胆酸(HA)来区分小儿氨酚黄那敏颗粒中体外培育牛黄和人工牛黄.采用Intersil ODS-3C18色谱柱,0.5%甲酸-乙腈(62∶38)为流动相,流速为1.0mL/min,柱温为40℃,进样量为20μL.采用蒸发散射光检测器,漂移管温度:110℃,载气为高纯氮气,流量:2.8L/min.HA保留时间为19.43min,检测限为2μg(浓度0.1mg/mL),含体外培育牛黄的小儿氨酚黄那敏颗粒未检出HA,含人工牛黄(0.2mg/mL)的小儿氨酚黄那敏颗粒可以检出HA.建立的方法可区分小儿氨酚黄那敏颗粒中的人工牛黄和体外培育牛黄,该法快速、简便、灵敏度高.
The HPLC-ELSD method is established for determining hyodeoxycholic acid(HA)to identify artificial and cultivated cow-bezoar in pediatric paracetamol,cow-bezoar and chlorphenamine maleate granules.The chromatographic conditions are as follows:Intersil ODS-3 C18(4.6 mm×250 mm,5μm)column;0.5%formic acid-acetonitrile(62∶38)as a mobile phase;flow rate at 1.0 mL/min;column temperature at 40℃;ELSD is used as a detector;drift tube temperature is 110℃and nitrogen flow rate is 2.8 L/min.The retention time of HA is 19.43 min.The limit of detection is 2μg(0.1 mg/mL).HPLC-ELSD can detect the existence of HA in pediatric paracetamol,cow-bezoar and chlorphenamine maleate granules with artificial cow-bezoar.However,HA could not be found in cultivated cow-bezoar(0.2 mg/mL).The method is rapid,simple,sensitive and suitable for the identification of artificial and cultivated cow-bezoar in pediatric paracetamol,cow-bezoar and chlorphenamine maleate granules.
