摘要
本文优化了蓖麻根系分泌有机酸的HPLC测定方法,检测采用PDA检测器,反向色谱柱InertSustain C18柱(4.6 mm×250 mm×5μm),检测波长为206 nm,柱温为30℃,流动相为0.01 mol/L KH2PO4,pH值为2.5,流速为0.3-0.8-0.3 mL/min变速。在该条件下检测蓖麻根系分泌的6种有机酸(草酸、苹果酸、乳酸、乙酸、柠檬酸和丙酸)分离效果好,用时短,分辨率高,灵敏度、准确度均能满足实际样品定量分析的要求,为研究铜离子胁迫影响蓖麻根系分泌有机酸的能力提供了快速有效的分析手段。
In this paper, a quantitative-analysis method with HPLC for the determination of organic acids secreted by the castor(Ricinus communis L.) root was optimized. The detection was performed with a PDA detector, and a reverse column InertSustain C18 column(4.6 mm×250 mm×5 μm) under the condition that the detection wavelength is 206 nm, the column temperature is 30 ℃, the mobile phase was 0.01 mol/L KH2 PO4, pH was 2.5. and the flow rate is 0.3-0.8-0.3 mL/min. In practical measurements, six organic acids secreted by castor root, including oxalic acid, malic acid, lactic acid, acetic acid, citric acid and propionic acid, were quantitatively analyzed with satisfactory measurement sensitivity and accuracy.
