超高效液相色谱-串联质谱法测定枸杞子中8种水溶性维生素 被引量:7

Determination contents of eight water-soluble vitamins in Lycii fructus by UPLC-MS/MS

摘要 建立同时测定枸杞子中核黄素、VB4、泛酸、烟酸、Vc、烟酰胺、吡哆醇、VB8等8种水溶性维生素的方法,并对宁夏、青海、甘肃、新疆等不同产地来源的枸杞子中该类成分进行含量评价。样品前处理用蒸馏水超声提取,采用色谱柱(2.1 mm×100 mm,1.8μm)分离,以10 mmol/L乙酸铵(A)-纯乙腈(B)为流动相梯度洗脱模式下,8种水溶性维生素在(0.0002~40)μg/m L浓度范围内线性良好,相关系数r2为0.9904~0.9996,方法精密度、重复性和稳定性的相对标准偏差(RSD)均小于2%,加样回收率为96.13%~102.15%,RSD值均小于3.50%。该研究建立的方法操作简便、准确、重复性好,可用于枸杞子中多种维生素的同步分析,为枸杞子营养价值测定及相关保健食品的开发提供了参考。 To establish a UPLC-MS/MS method for the simultaneous determination of eight water-soluble vitamins from Ningxia Hui Autonomous Region, Xinjiang Uygur Autonomous Region, Qinghai Province, Gansu Province of Lycii fructus, and analyze and investigate the difference of vitamin ingredients of Lycii fructus. After sample was treated with distilled water toultrasonic extraction, the separation of targeted compound was performed on a ACQUITY UPLC BEH C18 column(2.1 mm×100 mm, 1.8 μm) using 10 mmol/L ammonium acetate(A)-acetonitrile(B) as mobile phase under gradient elution mode. The linear range of 8 kinds of water-soluble vitamins was in the range of(0.0002~40)μg/mL with a correlation coefficient of 0.9904~0.9996. The precision, repeatability, stability and accuracy of the established method were well-validated and proved to be satisfactory, with the RSD<2%, the recovery rate was 96.13%~102.15%, and the RSD<3.80%. The method has the advantages of simple pretreatment, high sensitivity and reproducible, and is suitable for simultaneous analysis and determination of vitamins, and can provide reference for its health food development.
