云南香料烟中1个新黄酮及其抗菌活性

A new flavone from leaves of Nicotiana tabacum and its antibacterial activity

摘要 目的对云南香料烟的化学成分进行研究,试图从中发现新活性化合物。方法运用硅胶、凝胶及RP-HPLC等多种色谱技术进行分离纯化,并根据理化性质和波谱数据鉴定化合物的结构。结果从云南香料烟中分离得到2个烷基取代的黄酮类化合物,分别鉴定为2′-羟基-7-(2-羟乙基)-8-甲氧基-黄酮(1)和烟草黄酮A(2)。对化合物1和2进行了抗菌活性筛选,其对耐甲氧西林金黄色葡萄球菌(MRSA)菌株的MIC90值分别为(38±4)和(33±3)μg/m L。结论化合物1为新化合物,命名为香料烟黄酮A。化合物2首次从香料烟中分离到,化合物1和2均具有显著的抗菌活性。 Objective To fred the new biological compounds,the research of the chemical constituents in the leaves of Nicotiana tabacum had been carried out.Methods The chemical constituents in the leaves of N.tabacum were isolated by silica gel,Sephadex LH-20column chromatographies,and RP-HPLC methods.The structures of isolations were detected by using various spectroscopic techniques,including extensive 1D and 2D NMR techniques.Results Two C-alkylated fiavones were isolated from 95%ethanol extract of plant,which identified as 2'-hydroxy-7-(3-hydroxypropyl)-8-methoxy-fiavone (1)and tabaflavone A (2).Compounds 1and 2were evaluated for their anti-methicillin-resistant Staphylococcus aureus (anti-MRSA)activity.Conclusion Compound 2is isolated from the leaves of N.tabacum for the first time.Compound 1is a new compound named orientalflavone A.Compounds 1 and 2also show good anti-MRSA activity with MICg0value of (38±4)and (33±3)μg/mL,respectively.
