摘要
目的研究红毛七Caulophyllumrobustum的化学成分及其抗肿瘤活性。方法采用硅胶柱色谱、葡聚糖凝胶SephadexLH-20等色谱方法进行分离纯化,根据理化性质和波谱技术鉴定化合物的结构;采用CCK-8法,对化合物进行体外抑制人非小细胞肺癌A549细胞增殖的活性筛选。结果从红毛七根及根茎中分离出11个化合物,分别鉴定为collinsogenin(1)、常春藤皂苷元(2)、eechinocystic acid 3-O-α-L-arabinopyranoside(3)、magnolamide(4)、木兰箭毒碱(5)、columbamine(6)、4,4′-diphenylmethane-bis(methyl) Carbamate(7)、(-)-8,8′-二甲氧基-开环异落叶松树脂酚-L-O-β-D-吡喃葡萄糖苷(8)、紫花松果菊苷A(9)、2-[4-(3-hydroxy-1-propenyl)-2-methoxyphenoxy]-1,3-propanediol(10)、亚油酸甘油酯(11)。结论化合物1~2和4~11均首次从该属植物中分离得到;化合物1和2对人非小细胞肺癌A549细胞增殖具有中等强度的抑制作用。
Objective To study the chemical constituents of Caulophyllum robustum and its antitumor activities.Methods The chemical constittients were isolated and purified with silica gel column chromatography,dextran gel Sephadex LH-20,and other chromatographic methods.Their structures were determined by physicochemical and spectral analysis.Furthermore,the cytotoxicity of these chemical componerits against the NSCLC A549cell line was measured by CCK-8method.Results Eleven compounds were isolated and identified as collinsogenin (1),hederagenin (2),eechinocystic acid 3-O-α-L-arabinopyranoside (3),magnolamide (4), (-)-magnocurafine (5),columbamine (6),4,4'-diphenylmethane-bis (methyl)carbamate (7),(-)-8,8'-dimethoxy-L-O-(β-D- glucopyranosyl)secoisolariciresinol (8),asechipuroside A (9),2-[4-(3-hydroxy-l-propenyl)-2-methoxyphenoxy]-l,3-propanediol (10),and 1-1inoleoylglycerol (11).Conclusion Compounds 1-2and 4-11are isolated from Caulophyllum robustum genus for the first time.Compounds 1and 2show moderate inhibited effect on the proliferation of NSCLC A549cell line.
