摘要
目的:探究银屑平丸对BALB/c小鼠银屑病样模型血清中IL-6与IL-21表达的影响。方法:将30只BALB/c小鼠随机分为正常组、模型组、雷公藤组、银屑平丸高剂量组及低剂量组,每组6只。除正常组小鼠背部皮肤每日涂抹1次凡士林乳膏外,其余各组小鼠均每日外涂1次咪喹莫特乳膏,连续8 d。造模期间,各组小鼠同时进行灌胃给药:正常组与模型组每日用生理盐水灌胃1次,0.4 mL/次;雷公藤组每日用雷公藤多苷溶液药液灌胃1次,0.4 mL/次;银屑平丸高、低剂量组则每日分别用不同浓度的银屑平丸溶液药液灌胃1次,0.4 mL/次。每日给药24 h后观察各组小鼠背部皮肤的校正PASI评分;末次给药24 h后取血与皮肤组织。通过光镜下观察各组小鼠皮损组织形态学的改变,检测小鼠血清中IL-6与IL-21表达。结果:正常组的校正PASI评分趋势一直为0,其余各组的评分均为上升趋势。模型组、银屑平丸低剂量组、雷公藤组、银屑平丸高剂量组的PASI评分依次降低。模型组小鼠皮损组织为典型的银屑病样病理改变,除正常组小鼠无皮损外,其余各组小鼠均出现不同程度的角化过度及角化不全的病理改变。银屑平丸高剂量组小鼠血清IL-6、IL-21水平低于雷公藤组与银屑平丸低剂量组;雷公藤组小鼠血清IL-6、IL-21水平低于银屑平丸低剂量组。结论:银屑平丸可抑制BALB/c小鼠银屑病模型血清中IL-6与IL-21表达,这可能是银屑平丸治疗银屑病的作用机制之一。
Objective:To explore the influence of Yinxieping pills on serum IL-6 and IL-21 in the BALE/c mice psoriases model.Methods:A total of 30 BALB/c mice were randomLy divided into normal group,model group Leigongteng(雷公藤) group,Yinxieping pills high dose group and low dose group.Except that mice back skin of the normal group were coated with Vaseline cream once a day,the rest of the mice were coated with imiquimod cream daily for 8 days.During model establishment,all mice were intragastric administration on the same time:the normal group and the model group were treated with normal saline once a day by 0.4 mL/times;the Leigongteng group was treated with Leigongteng polygene solution by 0.4 mL/times;Yinxieping pills high and low dose groups were daily treated with different concentrations of Yinxieping pills solution by 0.4 mL/times.The corrected PASI score of the dorsal skin of each group was observed after 24 hours of the daily administration.Blood and skin tissue were collected after 24 hours of the last administration,and the results were finally obtained by light microscopy.The changes of IL-6 and IL-21 in the serum of mice were observed.Results:Except that the corrected PASI score trend of the normal group was always 0,the scores of the other groups arised.The PASI scores of model group,low dose group,Leigongteng group and high dose group were decreased in turn.The pathological changes of the psoriasis-like pathogens were observed in the model group.But the skin of the normal group unchanged.The mice skin in the other groups showed different degrees of hyperkeratosis and parakeratosis.Expression levels of IL-6 and IL-21 in the high dose group was lower than those in Leigongteng group and the Yinxieping pills low dose group,while theose in Leigongteng group was slower than those in the low dose group.Conclusion:It is one of the possible mechanisms that Yinxieping pills control psoriasis by inhibiting serum IL-6 and IL-21.
