补阳还五汤对多因素整合诱导“气虚血瘀"肺动脉高压大鼠的干预作用 被引量:3

The Effect of Buyang Huanwu Decoction(补阳还五汤) on Pulmonary Arterial Hypertension Rats with "Qi Deficiency and Blood Stasis Syndrome"

摘要 目的:观察多因素诱导"气虚血瘀"型肺动脉高压大鼠肺组织中的肺血流动力学及肺血管重建,探讨补阳还五汤治疗"气虚血瘀"型肺动脉高压的作用机制。方法:雄性Sprague-Dawley大鼠40只,随机分为空白对照组(C组)10只及造模组30只,均在SPF环境饲养;模型组进行"气虚血瘀"型肺动脉高压造模;再将造模组分为3组,每组10只;分别于制模后14 d,灌胃相应药物,连续用药15 d。肺动脉插管法测定平均肺动脉压(mPAP)、右心室收缩压(RVSP),计算右心肥厚指数(RVHI);处死动物后,观察各组大鼠肺组织病理改变并计算中膜厚度比(WT)和中膜面积比(WB)。结果:模型组大鼠肺动脉血管增厚,炎性浸润,WT,WB,mPAP,RVSP,RVHI高于空白对照组显著增加(P<0.01);补阳还五汤组及硝苯地平组与模型组比较,均能改善肺动脉血管增厚、炎性浸润,降低WT及WB,mPAP,RVSP,MAP,RVHI(P<0.01),但仍高于空白对照组(P<0.01)。结论:补阳还五汤可以改善肺动脉高压大鼠的肺血流动力学、右心室肥厚程度及肺组织病理改变,能部分逆转肺血管结构重建,有效降低肺动脉高压。 Objective:To observe the pulmonary hemodynamic and pulmonary vasculature remodeling within the lungs of multiple factors induced by "Qi deficiency and blood stasis syndrome" of in the lungs of pulmonary arterial hypertension rats with "Qi deficiency and blood stasis syndrome",and explore the mechanism of Buyang Huanwu decoction in the treatment of this disease.Methods:A total of 40 male Sprague-Dawley rats were randomly divided into control group(group C,10 rats) and modeling group(30 rats),keeping in the SPF environment.The model group was then divided into three groups,with10 rats in each group.On the 14 th day after the model establishment,the corresponding drugs were administered continuously for 15 days.The pulmonary artery intubation method was used to measure the mean pulmonary artery pressure(mPAP),right ventricular systolic pressure(RVSP),and the right heart hypertrophy index(RVHI).After the animals were sacrificed,the pathological changes of the lung tissue of each group were observed,and the median tunica thickness ratio(WT) and median tunica area ratio(WB) was calculated.Results:In the model group,pulmonary artery vascular thickening and inflammatory infiltration,WT,WB,mPAP,RVSP and RVHI were significantly increased when compared with the control group(P<0.01).As compared with the model group,the Buyang Huanwu decoction and nifedipine can improve pulmonary vascular thickening and inflammatory infiltration,and reduce WT and WB,mPAP,RVSP,MAP,RVHI(P<0.01).However,they were still higher than the normal control group(P<0.01).Conclusion:Buyang Huanwu decoction can improve pulmonary hemodynamics,right ventricular hypertrophy and pathological changes of lung tissue in rats with pulmonary arterial hypertension,which can partially reverse pulmonary vascular structural reconstruction and effectively reduce pulmonary arterial hypertension.
