杜仲叶总黄酮对围绝经期大鼠模型的影响 被引量:12

Effects of total flavonoids of Eucommia ulmoides leaf on the rat model of perimenopause

摘要 目的:观察杜仲叶总黄酮对不完全去势法制备围绝经期大鼠模型的影响。方法:各组分别给予大豆异黄酮、更年安以及大、中、小剂量杜仲叶总黄酮混悬液,连续给药30d,进行大鼠的Open field法测试,观察相关脏器指数,血清雌二醇(E2)、睾酮(T)、卵泡刺激素(FSH)、促黄体生成素(LH)、促性腺激素释放激素(GnRH)及骨钙素(BGP)、血浆中β-内啡肽(β-EP)的水平,相关组织的雌激素受体(ER)、雄激素受体(AR)含量。结果:与模型组比较,杜仲叶总黄酮能使大鼠活动距离和站立次数增加,使如胸腺指数、脾脏指数、子宫指数、肾上腺指数和血清E2、T、BGP、血浆β-EP水平提高,并使血清FSH、LH、GnRH水平降低;明显或显著的提高下丘脑、垂体、卵巢、子宫中ER水平,以及下丘脑和垂体中AR水平。结论:杜仲叶总黄酮不同剂量组可以对抗围绝经期综合征引起的性激素(E2、T、FSH、LH)水平紊乱,调整失调的雌雄激素受体(ER、AR)水平,纠正骨质疏松状态。 Objective:To observe the effects of total flavonoids of Eucommia ulmoides leaf on the preparation of perimenopause rat model.Methods:Each group was given soy isoflavones and Gengnian'an Pills,as well as large,medium and small dose of total flavonoids of Eucommia ulmoides leaf suspension liquid.Given the drug continuously for 30 days,the rats were tested by the Open field method,and the related viscera index,serum estradiol (E2),testosterone (T),follicle-stimulating hormone (FSH),luteinizing hormone (LH),gonadotrophin releasing hormone (GnRH)and osteocalcin (BGP),serum beta-endorphin (β- EP)levels,related organizations of estrogen receptor (ER)and androgen receptor (AR)content were observed.Results:Compared with the model group,the total flavonoids of Eucommia ulmoides leaf increased the distance of movement in rats,increased the levels of related organ indexes such as thymus,spleen,uterus and serum E2,T,BGP,and herpes-EP,and decreased the level of FSH,LH and GnRH in serum.It also significantly increased levels of ER in the hypothalamus,pituitary,ovary,uterus,and AR in the hypothalamus and pituitary.Conclusion:Different doses of total flavonoids of Eucomrnia ulmoides leaf can inhibit the level of sex hormones (E2,T,FSH,LH)caused by perimenopausaI syndrome,and adjust the level of androgen receptor (ER,AR)and rectify the osteoporosis status.
