摘要
以氯霉素半琥珀酸酯为母体直接偶联物固着在传感器末端形成具有双层结构的生物膜,通过检测端面生物膜层厚度变化所引起的光谱相位改变,实现样品中氯霉素的快速检测。结果表明,该传感器表现出良好的抗干扰性、重复性和稳定性,所建立的检测方法具有良好的灵敏度,不同样品基质检测对检测结果无显著影响。分别采用液相质谱和建立的金标记检测方法分析水性样品,其目标物检测结果一致,该方法检测下限为0.0125 ng/mL。生物膜干涉技术具有快捷、灵敏、特异性等特点,可在8 h内提供快速、特异的检测结果,尤适合大批量样品的现场快速筛查。
A double-layer structure biofilm achieved by chloramphenicol hemisuccinate conjugate immobilized at end-of-sensor was applied for rapid detecting the target concentration according to spectrum phase changes result from biofilm layer thickness change. The results showed that the established detection method was superior to the traditional immunoaffinity chromatographies with satisfying sensitivity, antiinterference, repeatability and stability. The aqueous samples were analyzed by liquid mass spectrometry and gold-labeled biofilm interferometry respectively with the detection results of the targets consistent. The detection limit of this method was 0.0125 ng/mL. Biofilm interference technology can be regarded as reliable and reproducible and is especially suitable for on-the-spot screening of large quantities of samples.
